Cost, multi-step protocols, equipment and radioactive safety requirements make this approach problematic in high-throughput testing or remote field use. We have developed an alternative simple, robust, inexpensive, one-step fluorescence assay method for antimalarial drug screening. Since non-parasitized erythrocytes lack DNA and RNA, a dye with DNA and RNA-induced fluorescence enhancement was selected to detect the presence of parasite nucleic acids. We evaluated the assay by determination of comparative IC50 values for known antimalarials against P. falciparum strain D6. Standard assay conditions were employed in 96-well plates including: RPMI-1640 with HEPES buffer, hypoxanthine, human serum and gentamicin; starting parasitemia was 2%. After 48 hours of growth, pre-mixed dye in lysis buffer was dispensed into each well, mixed and the plate was then left to incubate for 1 hour in the dark at room temperature. Fluorescence in each well was determined in a 96-well plate fluorometer, and these raw data used to calculate IC50. IC50 values for drugs including chloroquine, quinine, mefloquine, and artemisinin determined by this method were similar or identical to those obtained by standard radioisotope methods. Results from this new fluorescence assay suggest that it may be an ideal method for high-throughput drug screening, and that it could be modified for field use and faslodex.
