Results were observed in five neurons, and the average reduction in current amplitude in the presence of 50 100 M picrotoxin was 65 11%. Picrotoxin alone did not elicit any current. The observation that bumetanide and other loop diuretics have direct actions on GABAA receptors raises some concern about the interpretation of experimental results generated with these reagents. Furthermore, these inhibitors only selectively block NaK 2Cl cotransport over a limited concentration range and may have actions on other Cl transporters at drug concentrations that are required to block the cotransporter completely Russell, 2000 ; . Thus, it is important to use alternative experimental approaches to and butorphanol
The development of genetically altered mice has increased the use of mouse intestine in physiological research. The control data in this study are the first measures of active electrolyte transport in murine proximal colon. Together with our recent publication describing 17 ; the mouse distal colon, a more complete picture of electrolyte transport has emerged. Both proximal and distal colon exhibit a lumen-positive PD and net Na absorption when perfused with HCO3 Ringer. Like human, rat, and rabbit colon, the mouse colon has a proximal-to-distal gradient of conductance, although it appears to be somewhat more permeable than these tissues 3, 31 ; . Of interest is that mouse distal colon absorbs Cl at a greater rate than the proximal colon. It is unclear whether this reflects the experimental conditions or whether there are fewer or different Cl absorptive processes along the apical membranes of proximal colonic epithelial cells. The values for Na transport and various electrical parameters were similar in normal and GCC receptordeficient mice. However, net Cl absorption was lower in GCC ; mice than in normal mice, and a negative JR was observed indicating net HCO3 absorption ; . These differences most likely reflect a lower level of apical membrane Cl HCO3 exchange in receptor-deficient mice. Another possibility is that a basal HCO3 secretory process, normally active, is absent or reduced in GCC ; mice. Guanylin is secreted by mouse colonic mucosa, and active Cl and HCO3 secretion could be present in normal mice under basal conditions. However, if a basal anion secretory process is absent, JCl should be greater, not smaller, in GCC net ; mice than in GCC ; mice because of a lower Cl Js3m ; . Indeed, when basal secretion was tested for in normal mice with bumetanide and butyrate, no evidence for active, basal Cl secretion was found in either proximal or distal colon. Bumetanide inhibits Na -K 2Cl cotransport and thereby inhibits Cl entry across the basolateral membrane 1 ; . Butyrate appears to compete for apical anion channels in intestinal epithelia 10 ; . By the use of these same inhibitors, rat distal colon but not proximal colon ; exhibits active, basal Cl.

Vascular endothelial cells previously have been pmol K + g protein min. Na-K-C1 cotransport of aortic shown to possess a prominent Na-K-Cl cotransport endothelial cells is known to beregulated by a variety of system which mediates a K + influx of approximately vasoactive hormones and neurotransmitters and their second 20 pmol g of protein min. In the present nucleotides 3, 6 ; . This observation suggests that the promistudy we investigated the possibility that the high level nent endothelial cell cotransporter could play a role in mediof cotransport in these cells is due to a large number of effects of vasoactive agents on the vasculature. More Na-K-C1 cotransporters in the plasma membrane. This ating the was done by evaluating specific saturable binding of recently, evidence has been provided by this laboratory that Na-K-Cl cotransport may participate in regulation of endo[3H]bumetanide to cultured bovine aortic endothelial cells. We found a maximal r3H]bumetanide binding of thelial cell volume 7 ; . These findings indicate that Na-K-C1 0.83 pmol mg protein with a dissociation constant of cotransport may be of great importance to normal functioning 0.13 p ~ From these data, the number [3H]bumetan- of the endothelium of ide binding sites endothelial cell was determined to be In the present study, Na-K-Cl cotransport of endothelial approximately 230, 000, and the turnover number for cells was further investigated by evaluating the number of cell. cotransport activity was calculated to be K ions cotransporters present per endothelial The large magni300 site s. These findings indicatethat endothelial cells do tude of cotransport-mediated K' influx in thesecells could be a indeed exhibit large number of Na-K-Cl cotransport- the result of a large number of Na-K-Cl cotransporters presers cell relative to other cell types. We also investi- ent in the plasma membrane, a high turnover rate for each gated the effectson r3H]bumetanide binding of agents cotransporter, or a combination of both. In previous studies, known to modulate Na-K-Cl cotransport activity. Sat- saturable binding of [3H]bumetanide has been used successurable binding of [3H]bumetanide was found to be re- fully to quantitate the number of Na-K-C1 cotransporters duced significantly by treatment of the cells with 8- presentinmembranepreparations from dog, bovine, and bromo-cyclic AMP, 8-bromo-cyclic GMP, phorbol es- rabbit kidney, winter flounder intestine, shark rectal gland, 11 1 ters, norepinephrine, or rat atriopeptin , all of which and rabbit parotid gland 15-20 ; as well as in intact cells, have been shown to inhibit Na-K-Cl cotransport-me- including vascular smooth muscle cells, Ehrlich ascites tumor diated K + influx. cells, and duck and ferret erythrocytes 21-25 ; . Thus, in the present study, specific saturable [3H]bumetanide binding to intact bovine aortic endothelial cells was used to quantitate the numberof cotransporters present in the cells.
